Specificity and Use

ACTA2 / Smooth Muscle Actin antibody was raised against synthetic peptide: MCEEEDSTAL, corresponding to N terminal amino acids 1-10 of alpha smooth muscle Actin. Epitope: Acetyl group and the first 4 amino acids on the N-terminal end of the peptide chain of alpha-smooth actin. Percent identity by BLAST analysis: Human, Gorilla, Orangutan, Gibbon, Monkey, Marmoset, Mouse, Rat, Hamster, Elephant, Panda, Porcine, Turkey, Dog, Bat, Bovine, Horse, Rabbit, Chicken, Platypus (100%); Xenopus, Drosophila, Shewanella (80%).
Recognizes the alpha-smooth muscle isoform of actin. Does not react with actin from fibroblasts (b-and g-cytoplasmic), striated muscle (a-sarcomeric), and myocardium (a-myocardial). Species cross-reactivity: Cross-reacts with Human, Mouse, Rat, Rabbit, bovine, Baboon and Chicken.
Suitable for use in Immunofluorescence and Immunohistochemistry (Fr) (paraffin-embedded sections). Immunohistochemistry: Stains smooth muscle cells in vessel walls, gut walls, and myometrium. Myoepithelial cells in breast and salivary gland are also stained. Reacts with tumors arising from smooth muscles and myoepithelial cells. Positive control: Smooth muscle or leiomyosarcoma.

ACTA2 / Smooth Muscle Actin belongs to the actin family of proteins, which are highly conserved proteins that play a role in cell motility, structure and integrity. Alpha, beta and gamma actin isoforms have been identified, with alpha actins being a major constituent of the contractile apparatus, while beta and gamma actins are involved in the regulation of cell motility. This actin is an alpha actin that is found in skeletal muscle.
