Specificity and Use
MGMT antibody was raised against recombinant human MGMT (O6-methylguanine-DNA methyltransferase) expressed in E. coli.
Recognizes MGMT in human tissues, tumors, cells or xenograft lines. Weak cross-reactivity with MGMT from mouse, hamster and rat cells.
Suitable for use in Western Blot, Flow Cytometry, Immunohistochemistry and Immunoprecipitation and Immunofluorescence. Western Blot: 1:500 detects MGMT on 10 ug of Jurkat lysates. Immunohistochemistry: paraffin sections. Immunofluorescence (IC): HeLa cells. Positive control: Tonsil tissue, HeLa cells, CEM-CCRF cells.

Involved in the cellular defense against the biological effects of O6-methylguanine (O6-MeG) in DNA. Repairs alkylated guanine in DNA by stoichiometrically transferring the alkyl group at the O-6 position to a cysteine residue in the enzyme. This is a suicide reaction: the enzyme is irreversibly inactivated.
