Specificity and Use

CCL20 / MIP-3-Alpha antibody was raised against e. coli-derived recombinant human CCL20/MIP3-alpha Ala27-Met96 (Asn95-Asp) Accession # P78556.

Detects human CCL20/MIP3 alpha in direct ELISAs and Western blots. In direct ELISAs and Western blots, approximately 10% cross-reactivity.
Western Blot, 0.1 µg/ml, Recombinant Human CCL20/MIP-3 alpha. Immunocytochemistry, 5-15 µg/ml. Immunohistochemistry, 5-15 µg/ml. Neutralization, Measured by its ability to neutralize CCL20/MIP-3 alpha-induced chemotaxis in the BaF3 mouse pro-B cell line. transfected with human CCR6. The Neutralization Dose (ND 50)is typically 0.2-1.0 µg/ml in the presence of 10 ng/ml Recombinant Human CCL20/MIP-3 alpha.

C-terminal processed forms have been shown to be equally chemotactically active for leukocytes. Possesses antibacterial activity E.coli ATCC 25922 and S.aureus ATCC 29213.

CCL20/MIP3a i n Human Skin. CCL20/MIP3a was detected in immersion fixed paraffin-embedded sections of human skin using Goat Anti-Human CCL20/MIP3a Antigen Affinity-purified Polyclonal Antibody at 15 ug/ml overnight at 4°C. Tissue was stained using Anti-Goat HRP-DAB (brown) and counterstained with hematoxylin (blue).

CCL20/MIP-3 alpha in Human PBMCs. CCL20/MIP-3 alpha was detected in immersion fixed human peripheral blood mononuclear cells (PBMCs) stimulated with LPS using Human CCL20/MIP-3 alpha Antigen Affinity purified Polyclonal Antibody at 10 ug/ml for 3 hours at room temperature. Cells were stained using NorthernLights 557-conjugated Anti-Goat IgG Secondary Antibody (yellow) and counterstained with DAPI (blue).

Chemotaxis Induced by CCL20/MIP-3 alpha and Neutralization by Human CCL20/MIP-3 alpha Antibody. Recombinant Human CCL20/MIP-3 alpha chemoattracts the BaF3 mouse pro-B cell line transfected with human CCR6 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin. Chemotaxis elicited by Recombinant Human CCL20/MIP-3 alpha (10 ng/ml) is neutralized (green line) by increasing concentrations of Goat Anti-Human CCL20/MIP-3 alpha Antigen Affinity-purified Polyclonal Antibody. The ND50 is typically 0.2-1.0 ug/ml.
