Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Description
SMAD1 antibody LS-C529401 is a biotin-conjugated rabbit polyclonal antibody to SMAD1 from human, mouse and pig. Validated for WB.
Target
Human SMAD1
Host
Rabbit
Reactivity
Human, Mouse, Pig (tested or 100% immunogen sequence identity)
Clonality
IgG Polyclonal
Conjugations
Biotin. Also available Unconjugated or conjugated with AP, APC, FITC, HRP, PE.
Purification
Immunoaffinity purified
Modifications
Unmodified
Applications
  • Western blot
  • (applications tested for the base form of this product only)
Immunogen
SMAD1 antibody was raised against synthetic peptide derived from an internal region of the human Smad1 protein.
Specificity
Reacts with the human, mouse and pig Smad1 proteins. On Western blots, it identifies a single band at ~52kD. A ~140kD band of unknown origin is seen in SK-OV-3 cell lysates and a ~200kD band of unknown origin is seen in NIH3T3 cell lysates. Reactivity has been confirmed with human HepG2 and SK-OV-3, African Green monkey COS-7, and mouse NIH3T3 cell lysates, as well as with fetal mouse muscle and thymus homogenates.
Presentation
PBS, pH 7.2, No preservatives added
Storage
Store at 4°C.

Applications: ICC, Western blot, Immunoprecipitation, Flow Cytometry
Anti-SMAD1 Antibody - Western Blot. Western blot of Affinity Purified anti-SMAD1 antibody shows detection of endogenous SMAD1 in whole cell lysates from human hepatoma (HEPG2, lanes 1-4) and keratinocyte (HaCaT, lanes 5-8) derived cell lines treated with PBS, BMP2, EGF, or NaCl for 1 h at 37?C before harvest. Each lane contains approximately 15 ug of lysate. Primary antibody was used at a 1:500 dilution in 1% BLOTTO (B501-0500) and reacted for 1 hour at room temperature. Anti-beta actin (p/n LS-B323) staining was used as a loading control. The membrane was washed and reacted with a 1:3000 dilution HRP-conjugated a-Rabbit IgG (p/n LS-C60865) for 1 hour at room temperature. Detection was by ECL. Personal communication, Xin-Hua Feng, Baylor College of Medicine, Houston, TX.
Species: Human, Mouse, Rat, Dog

Applications: Western blot
Immunofluorescent staining using SMAD1 antibody. Immunostaining analysis in HeLa cells. HeLa cells were fixed with 4% paraformaldehyde and permeabilized with 0.01% Triton-X100 in PBS. The cells were immunostained with anti-SMAD1 antibody.
