RAF1 / RAF antibody was raised against 12 residue synthetic C terminal peptide (CTLTTSPRLPVF) corresponding to residues 637-648 of human Raf-1. Percent identity by BLAST analysis: Human, Gorilla, Orangutan, Gibbon, Monkey, Marmoset, Mouse, Rat, Hamster, Elephant, Panda, Dog, Bat, Bovine, Horse, Rabbit (100%).
Recognizes Raf-1, Mr ~70kD. Species cross-reactivity: Human. Predicted to cross-react with mouse and rat based on sequence homology.
Suitable for use in Western Blot, Immunoprecipitation. Western Blot Analysis: 2 ug/ml detects Raf-1 in RIPA lysates from 3T3/A31 cell lysates. 3T3/A31 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Raf-1 (2 ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Included Positive Antigen Control: 3T3/A31 cell lysate. Add 2.5 ul of 2-mercaptoethanol/100 ul of lysate and boil for 5 minutes to reduce the preparation. Load 20 ug of reduced lysate per lane for minigels. MAPK Activation and MBP Phosphorylation: 4 ug immunoprecipitates Raf-1. The immunocomplex was then used in a coupled phosphorylation assay to activate MEK1, which in turn activates MAP Kinase 2 that phosphorylates myelin basic protein in vitro.
