Specificity and Use

TP53BP1 / 53BP1 antibody was raised against a synthetic peptide, which represented a portion of human tumor protein p53 binding protein 1 encoded exon 19 (LocusLink ID 7158).
Specific for human 53BP1 protein. Species cross-reactivity: Reacts with human in Western Blot and immunocytochemistry/immunofluorescence and it reacts with mouse in immunocytochemistry/immunofluorescence (Western Blot has not been done using this antibody
Suitable for use in Immunofluorescence, Western Blot, Immunohistochemistry (paraffin-embedded sections) and Immunocytochemistry. Immunofluorescence: 1:200-1:1000. Western Blot: 1:2000-1:10000. A band at ~220kD is seen on SDS-PAGE. Immunocytochemistry: 1:200-1:1000. Positive control: U205, 293T, and MO59K cell lysates (human) and MEF lysates (mouse).

Tumor protein p53 binding protein 1 binds to the central domain of p53 required for site-specific DNA binding thus inhibiting the ability of p53 to bind DNA. TP53BP1 also functions as a checkpoint protein with properties of a DNA double-strand break sensor. It is a putative substrate of ATM kinase and is phosphorylated in response to DNA damage and localizes to sites of double-strand breaks.
