Specificity and Use
SMAD3 antibody was raised against a 20 amino acid synthetic peptide derived from a central portion of the linker domain of human Smad3.
Recognizes human Smad3. The antibody shows very minor cross-reactivity with Smad2 (using relatively high concentrations of recombinant Smad2).
Suitable for use in ELISA, Immunohistochemistry and Western Blot. ELISA: 0.1-1 ug/ml. Western Blot: 1-3 ug/ml. Immunohistochemistry: 1 ug/ml Requires HIER with EDTA, pH 8.0 prior to staining. Positive control: Western Blot: human Smad3-transfected COS.

Receptor-regulated SMAD (R-SMAD) that is an intracellular signal transducer and transcriptional modulator activated by TGF-beta (transforming growth factor) and activin type 1 receptor kinases. Binds the TRE element in the promoter region of many genes that are regulated by TGF-beta and, on formation of the SMAD3/SMAD4 complex, activates transcription. Also can form a SMAD3/SMAD4/JUN/FOS complex at the AP-1/SMAD site to regulate TGF-beta-mediated transcription.
