MCAM / CD146 antibody was raised against human primary WM902B melanoma cells.
Recognizes MEL-CAM, also known as MUC18, CD146, S-Endo-1, or A32 melanomaassociated antigen, Mr ~113kD. Species cross-reactivity: Human specific.
Suitable for use in Western Blot, Immunohistochemistry, Immunoprecipitation and Immunocytochemistry. Western Blot: 1-2 ug/ml detected MEL-CAM lysates from SK-Mel-37 melanoma cell lysates. SK-Mel-37 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-MEL-CAM (MUC18, CD146) (1 ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunohistochemistry: Detects MEL-CAM in human melanoma samples by PAP immunohistochemistry. Immunoprecipitation: Reported by an independent laboratory to immunoprecipitate MEL-CAM from WM1366 melanoma cell lysates. Immunocytochemistry: Detects MEL-CAM in human keratinocytes.

Plays a role in cell adhesion, and in cohesion of the endothelial monolayer at intercellular junctions in vascular tissue. Its expression may allow melanoma cells to interact with cellular elements of the vascular system, thereby enhancing hematogeneous tumor spread. Could be an adhesion molecule active in neural crest cells during embryonic development.
