Specificity and Use

GNAS antibody was raised against synthetic peptide corresponding to the C-terminal region of bovine Gs alpha (RMHLRQYELL) (KLH). Percent identity by BLAST analysis: Human, Gorilla, Gibbon, Monkey, Marmoset, Mouse, Rat, Hamster, Elephant, Panda, Bat, Bovine, Dog, Horse, Pig, Opossum, Turkey, Chicken, Platypus, Xenopus, Seabass, Stickleback, Pufferfish, Zebrafish, Drosophila, Mosquito (100%).

Recognizes both the large (MW 48-52kD) and small (MW43-45kD) forms of bovine Gs alpha. Does not cross-react with other G proteins.
Western Blot: 1:1000-1:2000 detected both forms of Gs alpha as a doublet in 20ug of rat brain microsomal preparation. Rat brain microsomal preparation was resolved by electrophoresis, transferred to nitrocellulose and probed with LS-C24097 (1:1000). Proteins were visualized using a IgG (HRP) goat anti-rabbit and a chemiluminescence detection system.Immunoprecipitation: 2ul immunoprecipitates Gs alpha(45kD) from 500ug of solubilized CHO and mouse brain membrane preparations.Immunocytochemistry: 1:200 detected Gs alpha in 3T3 cells fixed for 3 minutes with 100% ice-cold acetone.Optimal dilutions to be determined by researcher.

The GNAS1 gene encodes the alpha subunit of the G protein Gs, which couples receptor binding by several hormones to activation of adenylate cyclase (Hayward et al., 1998). This locus has a highly complex imprinted expression pattern. It gives rise to maternally, paternally, and biallelically expressed transcripts that are derived from four alternative promoters and 5' exons.
