Specificity and Use

CD274 / B7-H1 / PD-L1 antibody was raised against purified, NS0-derived, recombinant mouse B7 homolog 1 (rmB7-H1) extracellular domain.
Mouse B7-H1< 0.01 EU per 1 ug of the antibody as determined by the LAL method.
Direct ELISA-This antibody can be used at 0.5-1.0 µg/ml with the appropriate secondary reagents to detect mouse B7-H1. The detection limit for rmB7-H1 is approximately 0.3 ng/well. In this format, this antibody shows approximately 20% cross-reactivity with rhB7-H1. Western blot-This antibody can be used at 0.1-0.2 µg/ml with the appropriate secondary reagents to detect mouse B7-H1. The detection limit for rmB7-H1 is approximately 5 ng/lane under non-reducing and reducing conditions, respectively. Immunohistochemistry-This antibody will detect B7-H1 in cells and tissues. The working dilution is 2-15 µg/ml. Flow Cytometry-This antibody can be used at 3-10 µg/106 cells with an appropriate secondary antibody for indirect immunofluorescence staining of cells by flow cytometry.

CD274, also known as B7-H1 and PD-L1, a cell surface glycoprotein, is a member of the B7 family of co-stimulatory molecules. CD274 is expressed constitutively on macrophages and dendritic cells, and is induced on activated T-cells, B-cells, endothelial cells and epithelial cells in response to interferons. CD274 has dual functions: inhibition of activated effector T cells and co-stimulation of naïve T cells.
