Specificity and Use

CCND1 / Cyclin D1 antibody was raised against synthetic peptide corresponding to the 11 C-terminal amino acids (residues 285-295; sequence: CTPTDVRDVDI) of human Cyclin D1. Percent identity by BLAST analysis: Human, Gorilla, Orangutan, Gibbon, Monkey, Mouse, Rat, Hamster, Bat (100%); Elephant, Panda, Bovine, Dog, Horse, Turkey, Chicken, Platypus, Lizard, Xenopus, Salmon, Pufferfish (91%); Opossum, Stickleback, Zebrafish (82%).

Recognizes p36 Cyclin D1 and cross-reacts with p34 Cyclin D2, which has a highly homologous epitope. No cross-reactivity with Cyclin A or Cyclin B. Shows reactivity with a 28kD band that can competed away by pre-incubating antibody with the peptide immunogen. Also detect a nonspecific band at 21kD. Species cross-reactivity: mouse and rat.
Western Blot Analysis 2: At 0.5-2 ug/ml, this antibody detected Cyclin D in a RIPA lysates of human A431 cells, mouse 3T3 fibroblasts. Positive Antigen Control: EGF-stimulated A431 cell lysate. Use 20 ug per lane. Immunoprecipitation: 5 ug of antibody immunoprecipitated Cyclin D from a RIPA lysate of human A431 cells. An H1 kinase assay can be performed with the immunoprecipitate, as reported previously. Immunocytochemistry: At 10 ug/ml, a previous lot of antibody detected Cyclin D in cells fixed with 4% paraformaldehyde/0.1% Triton X-100. Western Blot Analysis EGF-stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Cyclin D (2 ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.

Regulatory component of the cyclin D1-CDK4 (DC) complex that phosphorylates and inhibits members of the retinoblastoma (RB) protein family including RB1 and regulates the cell-cycle during G1/S transition. Phosphorylation of RB1 allows dissociation of the transcription factor E2F from the RB/E2F complex and the subsequent transcription of E2F target genes which are responsible for the progression through the G1 phase. Hypophosphorylates RB1 in early G1 phase.
