LS-F40010 is a 96-well enzyme-linked immunosorbent assay (ELISA) for the Quantitative detection of Mouse CDKN3 / KAP in samples of Plasma, Serum and Tissue Homogenates. It is based upon a Competitive EIA assay principle and can be used to detect levels of CDKN3 / KAP as low as 0.094 nanograms per millilter.

This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with Kap. During the reaction, Kap in the sample or standard competes with a fixed amount of Kap on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to Kap. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin(SABC) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of Kap in the samples is then determined by comparing the O.D. of the samples to the standard curve.
CDKN3 encodes a dual specificity protein phosphatase which has been identified as a cyclin-dependent kinase inhibitor and has been shown to interact with, and dephosphorylate human CDK2 kinase. CDKN3 has also been shown to associate with human CDK3 and CDC2. The protein has been reported to remove phosphate from tyrosine residues in model substrates in vitro; however, a mutant protein that bears a lesion in the putative active site cysteine has been shown to lose catalytic activity.
