Specificity and Use

ZIC2 antibody was raised against synthetic peptide C-HSGLSSNFNEWY from the C-terminus of human ZIC2 (NP_009060.2). Percent identity by BLAST analysis: Human, Chimpanzee, Gorilla, Orangutan, Gibbon, Monkey, Marmoset, Mouse, Rat, Bat, Elephant, Opossum, Guinea pig (100%); Galago, Tamarin, Bovine, Dog, Hamster, Panda, Porcine, Xenopus, Medaka, Seabass, Pufferfish, Zebrafish, Stickleback (92%); Zebra finch, Chicken, Lizard, Salmon (83%).

Human ZIC2. This antibody is expected to NOT cross-react with other ZIC proteins.
Peptide ELISA: antibody detection limit dilution 1:1000. Western blot: In transfected RWPE1 transiently expressing Human ZIC2-Myc a band of approx. 65kD is observed. This band is not observed in the non-transfected RWPE1, nor when GFP is transiently expressed. The calculated molecular size is 55.0kD according to NP_009060.2 and 58.6kD for the ZIC2-Myc protein. Data provided by Nathan Bowen and Maryam Boseman, Clark Atlanta University, GA, USA. We have observed an approx 75kD band in Human Brain (Cerebellum and Cerebral Cortex) lysates Recommended concentration, 0.5-2 ug/ml.

Acts as a transcriptional activator or repressor. Plays important roles in the early stage of organogenesis of the CNS. Activates the transcription of the serotonin transporter SERT in uncrossed ipsilateral retinal ganglion cells (iRGCs) to refine eye-specific projections in primary visual targets. Its transcriptional activity is repressed by MDFIC. Involved in the formation of the ipsilateral retinal projection at the optic chiasm midline.

ZIC2 antibody RWPE1 lysate (60 ug protein in RIPA buffer) overexpressing Human ZIC2 with C-terminal MYC tag probed with (0.5 ug/ml) in the right panel and probed with anti-MYC Tag (1/1000) and anti-beta-Actin in the left panel. Mock-transfected RWPE1 in lanes 1 and expressing GFP in lanes 2. Primary incubations were overnight at 4C. Detected by chemiluminescence.
