Specificity and Use

WNT1 antibody was raised against synthetic peptide corresponding to an internal region of human Wnt1 protein.
human and mouse Wnt1 protein. A BLAST analysis was used to suggest cross-reactivity with Wnt1 from mouse, human, rat, bovine, dog, macaque, opossum and rat based on a 100% homology with the immunizing sequence. Partial cross-reactivity is expected against chicken Wnt1 based on a 91 % sequence homology. Cross-reactivity with Wnt1 from other sources has not been determined
This affinity purified antibody has been tested for use in ELISA and western blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately ~41 kD in size corresponding to Wnt1 by western blotting in the appropriate cell lysate or extract. This antibody is capable of detecting endogenous Wnt1.

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It is very conserved in evolution, and the protein encoded by this gene is known to be 98% identical to the mouse Wnt1 protein at the amino acid level.

Anti-Wnt1 Antibody - Western Blot. Western blot of affinity purified anti-Wnt1 antibody shows detection of endogenous Wnt1 in human-derived MCF7 cell lysate (lane 1) and mouse-derived 3T3 cell lysate (lane 2). The band at ~41 kD, indicated by the arrowhead, corresponds to Wnt1. After transfer, the membrane was blocked with 5% BLOTTO. Primary antibody was used at a 1:1400 dilution in PBS containing 1% BLOTTO. The specificity of the antibody was confirmed by peptide competition which completely blocked reaction of the antibody with Wnt1 (data not shown).
