Specificity and Use

TP73 / p73 antibody was raised against a synthetic peptide C-THSPYAQPSS corresponding to amino acids 95-104 of human p73, GenBank EAW71468.1. This peptide sequence is 100% conserved across multiple p73 isoforms and between rat, mouse, dog and bovine p73. Percent identity by BLAST analysis: Human, Gorilla, Gibbon, Monkey, Marmoset, Mouse, Rat, Hamster, Elephant, Panda, Dog, Bovine (100%); Opossum, Chicken, Xenopus (90%); Bat, Rabbit, Turkey, Stickleback, Medaka, Pufferfish, Zebrafish, Nectri, Fusarium (80%).

This peptide sequence is 100% conserved across multiple p73 isoforms and between rat, mouse, dog and bovine p73. Researchers are encourage to use the the immunogen sequence information and available Bioinformatics data bases to obtain additional information regarding the amino acid sizes associated with various p73 isoforms.
Immunohistochemistry: LS-B2084 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2084 was determined to be 5 ug/ml.

This gene encodes tumor protein p73, which is a member of the p53 family of transcription factors involved in cellular responses to stress and development. The family members include p53, p63, and p73 and have high sequence similarity to one another, which allows p63 and p73 to transactivate p53-responsive genes causing cell cycle arrest and apoptosis.
