Specificity and Use

TGFB1 / TGF Beta 1 antibody was raised against synthetic peptide from human TGFB1 / TGF Beta.
aa286-293
Aa 286-293 (of 390) of full-length (including signal peptide) precursor TGFb protein. This sequence also corresponds with aa 8-15 (of 112) of the mature growth factor.
Immunohistochemistry: LS-B655 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B655 was determined to be 5 ug/ml.

TGF Beta is a multifunctional peptide that controls proliferation, differentiation, and other functions in many cell types. TGFB acts synergistically with TGFA in inducing transformation. It also acts as a negative autocrine growth factor. Dysregulation of TGFB activation and signaling may result in apoptosis. Many cells synthesize TGFB and almost all of them have specific receptors for this peptide. It plays an important role in bone remodeling.

Western blot
Each lane contains 250 ng of protein under non-reducing (lane 1) and reducing conditions (lane 2). The blot was incubated with a 1:200 dilution of the antibody at room temperature for 1 h followed by detection using IRDye 800 labeled Goat-a-Rabbit IgG (H&L). IRDye 800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR.
