Specificity and Use
PMEL / SILV / gp100 antibody was raised against melanoma Associated Antigen (100/7).
Recognizes a human (pre) melanosomal 100+7kD antigen (glycoprotein). Reacts with melanomas, clear cells sarcomas (melanoma of soft tissue), nevocellular nevi and normal melanocytes. Except for one case of non-Hodgkin's lymphoma in which macrophages were positive, no reactions with other tumors or tissues have been observed.
Immunohistochemistry (Immunoperoxidase staining on frozen tissue sections): 1:2.5-1:10 with PBS.
PBS, pH 7.4, 0.1% BSA, 0.02% sodium azide.

PMEL / SILV / gp100 is a melanocyte-specific type I transmembrane glycoprotein. The encoded protein is enriched in melanosomes, which are the melanin-producing organelles in melanocytes, and plays an essential role in the structural organization of premelanosomes. This protein is involved in generating internal matrix fibers that define the transition from Stage I to Stage II melanosomes.
