Specificity and Use

P2RY1 / P2Y1 antibody was raised against synthetic peptide C-RSEANLQSKSED from the C-terminus of human P2RY1 / P2Y1 (NP_002554.1). Percent identity by BLAST analysis: Human, Gorilla, Gibbon, Monkey, Marmoset, Elephant, Panda, Bat, Dog, Bovine, Rabbit, Opossum, Guinea pig (100%); Mouse, Rat, Hamster, Horse (92%); Lizard, Xenopus (83%).

Peptide ELISA: antibody detection limit dilution 1:8000. Western Blot: Preliminary experiments gave an approx. 50kD band in lysates of Human Lung and of cell line A549 after 0.3 ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 42.1kD according to NP_002554.1. The 50kD band was successfully blocked by incubation with the immunizing peptide. We would appreciate any feedback from people in the field - have any results been reported with other antibodies / lysates? Have any further splice variants / modified forms been reported?

P2RY1 / P2Y1 belongs to the family of G-protein coupled receptors. This family has several receptor subtypes with different pharmacological selectivity, which overlaps in some cases, for various adenosine and uridine nucleotides. This receptor functions as a receptor for extracellular ATP and ADP. In platelets binding to ADP leads to mobilization of intracellular calcium ions via activation of phospholipase C, a change in platelet shape, and probably to platelet aggregation.
