PBMC: Add 10 ul of antibody/10^6 PBMC in 100 ul PBS. Mix gently and incubate for 15 minutes at 2° to 8o C. Wash twice with PBS and analyze. WHOLE BLOOD: Add 10 ul of antibody/100 ul of whole blood. Mix gently and incubate for 15 minutes at room temperature 20o C. Lyse the whole blood. Wash once with PBS and analyze. See instrument manufacturers instructions for Lysed Whole Blood and Immunofluorescence analysis with a flow cytometer or microscope.

MME / CD10 is a common acute lymphocytic leukemia antigen that is an important cell surface marker in the diagnosis of human acute lymphocytic leukemia (ALL). This protein is present on leukemic cells of pre-B phenotype, which represent 85% of cases of ALL. This protein is not restricted to leukemic cells, however, and is found on a variety of normal tissues.
