Plays an essential role in the regulation of angiogenesis, vascular development, vascular permeability, and embryonic hematopoiesis. Promotes proliferation, survival, migration and differentiation of endothelial cells. Promotes reorganization of the actin cytoskeleton. Isoforms lacking a transmembrane domain, such as isoform 2 and isoform 3, may function as decoy receptors for VEGFA, VEGFC and/or VEGFD.
Phospho-VEGF R2/KDR/Flk-1 in A431 Human Cell Line. VEGF R2/KDR/Flk-1 phosphorylated at Y1214 was detected in immersion fixed A431 human epithelial carcinoma cell line untreated (lower panel) or treated (upper panel) with pervanadate using Rabbit Anti-Human Phospho-VEGF R2/KDR/Flk-1 Antigen Affinity-purified Polyclonal Antibody at 10 ug/ml for 3 hours at room temperature. Cells were stained using the NorthernLights 557-conjugated Anti-Rabbit IgG Secondary Antibody (red) and counterstained with DAPI (blue).
Detection of Human Phospho VEGF R2/KDR/Flk-1 by Western Blot. Western blot shows lysates of HUVEC human umbilical vein endothelial cells untreated(-) or treated (+) with 100 uM pervanadate (PV) for 10 minutes. PVDF membrane was probed with 0.5 ug/ml of Rabbit Anti-Human Phospho-VEGF R2/KDR/Flk-1 Antigen Affinity-purified Polyclonal Antibody, followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody. Specific bands were detected for Phospho-VEGF R2/KDR/ Flk-1 at approximately 210 kD (as indicated). This experiment was conducted under reducing conditions.
Detection of Phospho VEGF R2/ KDR/Flk1 in pervanadate-treated HUVEC by Flow Cytometry. HUVEC human umbilical vein endothelial cells were unstimulated (yellow filled histogram) or treated with 100 uM pervanadate for 15 minutes (orange filled histogram), then stained with Rabbit Anti-Human Phospho VEGF R2/KDR/Flk1 Antigen Affinity-purified Polyclonal Antibody, or control antibody (open histogram), followed by Phycoerythrin-conjugated Anti-Rabbit IgG Secondary Antibody. To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with methanol.
