Specificity and Use

IL17D antibody was raised against e. coli-derived recombinant human Interleukin 27 (aa18-202).
Recognizes human IL-27. Does not cross-react with rhIL-17, rhIL17B, rhIL-17C, rhIL-17E, or rhIL-17F. Species sequence homology: Mouse: 86%.
Suitable for use in Western Blot, Flow Cytometry, Immunohistochemistry and ELISA. Western Blot: 1-2 ug/ml. Detection limit is approximately 25 ng/lane under non-reducing conditions. Reducing conditions are not recommended. Flow Cytometry: 25 ug/ml. For intracellular staining, cells must be fixed and permeabilized using 4% paraformaldehyde and 0.1% saponin in PBS. Add 10 ul of the diluted solution to 1-5x10^5 cells in a total reaction volume not exceeding 200ul. Incubate for 30 minutes and then wash cells with 0.1% saponin prior to addition of a secondary developing reagent. Cells should be washed a final time in 0.1% saponin prior to flow cytometric analysis. Direct ELISA: 0.5-1 ug/ml. Detection limit is ~5 ng/well. Immunohistochemistry: 8-25 ug/ml with appropriate secondary reagents to detect IL-17D in paraffin-embedded normal heart tissue sections.


Lyophilized from PBS, 5% trehalose.
200 µl Reconstitute with sterile 40-50% glycerol, PBS.
Store at -20°C lyophilized. Once reconstituted, aliquot and store at -20°C.

IL17D is a cytokine that shares the sequence similarity with IL17. The treatment of endothelial cells with this cytokine has been shown to stimulate the production of other cytokines including IL6, IL8 and CSF2/ GM-CSF. The increased expression of IL8 induced by this cytokine was found to be NF-kappa B-dependent.
