Specificity and Use

CD110 / MPL antibody was raised against synthetic peptide corresponding to amino acids 615-628 of human TPO (thrombopoietin) receptor [C-ANHSYLPLSYWQQP] (KLH-conjugated). Species sequence Homology: Mouse - 100%. Percent identity by BLAST analysis: Human, Monkey, Mouse, Hamster (100%); Elephant, Rabbit (93%); Panda, Bovine, Pig (86%).
Recognizes glycosylated and unglycosylated human TPO receptor/c-Mpl, Mr 70kD. Species cross-reactivity: mouse. Species sequence Homology: mouse - 100%.
Suitable for use in Western Blot, Immunohistochemistry and Immunoprecipitation. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunohistochemistry: Stains megakaryocytes in bone marrow biopsy sections. Immunoprecipitation: 5 ug immunoprecipitates TPO receptor from 500 ug of DAMI RIPA lysate.

In 1990 an oncogene, v-mpl, was identified from the murine myeloproliferative leukemia virus that was capable of immortalizing bone marrow hematopoietic cells from different lineages. In 1992 the human homologue, named, c-mpl, was cloned. Sequence data revealed that c-mpl encoded a protein that was homologous with members of the hematopoietic receptor superfamily. Presence of anti-sense oligodeoxynucleotides of c-mpl inhibited megakaryocyte colony formation.
