The scaffolding protein encoded by this gene is the main component of the caveolae plasma membranes found in most cell types. The protein links integrin subunits to the tyrosine kinase FYN, an initiating step in coupling integrins to the Ras-ERK pathway and promoting cell cycle progression. The gene is a tumor suppressor gene candidate and a negative regulator of the Ras-p42/44 mitogen-activated kinase cascade.
PhosphoCaveolin1 (Y14 ) in MCF7 Human Cell Line. Caveolin1 phosphorylated at Y14 was detected in immersion fixed MCF7 human breast cancer cell line using Mouse Anti-Human PhosphoCaveolin1 (Y14) Monoclonal Antibody at 25 ug/ml for 3 hours at room temperature. Cells were stained using the NorthernLights 557-conjugated Anti-Mouse IgG Secondary Antibody (yellow) and counterstained with DAPI (blue). Specific staining was localized to plasma membranes.
Detection of Human PhosphoCaveolin1 (Y14) by Western Blot. Western blot shows lysates of HUVEC human umbilical vein endothelial cells and A431 human epithelial carcinoma cell line untreated () or treated (+) with 1 mM Pervanadate (PV) for 5 minutes. PVDF membrane was probed with 0.2 ug/ml of Mouse Anti-Human PhosphoCaveolin1 (Y14) Monoclonal Antibody followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody. A specific band was detected for PhosphoCaveolin1 (Y14) at approximately 22 kD (as indicated). This experiment was conducted under reducing conditions and using Western Blot Buffer Group 1.
