Specificity and Use

AKT1 antibody was raised against sf 21-derived recombinant human v-akt murine thymoma viral oncogene homolog 1 aa2-480.

Recognizes human Akt1. Does not detect rhAkt2 or rhAkt3.
Suitable for use in ELISA, Western Blot, Immunocytochemistry, Suitable for use in Flow cytometry: 50 ug/ml. This antibody was validated for flow cytometry using MCF-7 cells. For. intracellular staining to detect Akt1, cells must first be fixed and permeabilized using 4% paraformaldehyde and 0.1% saponin in PBS. Dilute this antibody to 50 ug/ml and add 10 ul of the diluted solution to 1-5x10^5 cells in a total reaction volume not exceeding 20ulL. The binding of unlabeled monoclonal antibodies may be visualized by adding a secondary developing reagent such as goat anti-mouse IgG conjugated to a fluorochrome. Cells should be washed for a final time in 0.1% saponin prior to flow cytometric analysis. Immunocytochemistry: 8-25 ug/ml to detect Akt1 in fixed human peripheral blood mononuclear cells.

AKT1 is one of 3 closely related serine/threonine-protein kinases (AKT1, AKT2 and AKT3) called the AKT kinase, and which regulate many processes including metabolism, proliferation, cell survival, growth and angiogenesis. Over 100 substrate candidates have been reported so far, but for most of them, no isoform specificity has been reported.
