solute carrier family 26 (anion exchanger), member 4

Mutations in this gene are associated with Pendred syndrome, the most common form of syndromic deafness, an autosomal-recessive disease. It is highly homologous to the SLC26A3 gene; they have similar genomic structures and this gene is located 3' of the SLC26A3 gene. The encoded protein has homology to sulfate transporters.

SLC26A4 / Pendrin Antibody - Immunohistochemistry image at a dilution of 1:500 and staining in paraffin-embedded human melanoma cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0) . Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4 °C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

SLC26A4 / Pendrin Antibody - Western blot analysis of SLC26A4 using anti-SLC26A4 antibody. Lane 1: 50 µg human HK-2 whole cell lysates, Lane 2: 50 µg human 293T whole cell lysates. Electrophoresis was performed on a 5-20% SDS-PAGE under reducing conditions. Transferred to a Nitrocellulose membrane. Blocked with 5% Non-fat Milk/ TBS for 1.5 hour at RT. Incubated with anti-SLC26A4 polyclonal antibody at 0.5 µg/mL overnight at 4°C. Washed with TBS-0.1%Tween 3 times with 5 minutes each. Incubated with goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. Signal was developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for SLC26A4 at approximately 86KD. The expected band size for SLC26A4 is at 86KD.
