Myotubularin-related protein 5 (MTMR5), a myotubularin-related phosphatase, has been shown to be similar to dual-specificity phosphatases of the myotubularin group; however MTMR5 also has been shown to lack several crucial residues in the catalytic pocket which render it catalytically inactive as a phosphatase. The pocket has been shown to bind phosphorylated synthetic substrates which suggests that it may be a protective factor that competes with functional phosphatases for substrate interaction. MTMR5 also has been shown to bind the phosphoinositides PtdIns(3,4)P2 and PtdIns(3,4,5)P3 in a PH (pleckstrin homology) domain. Also known as SET-binding factor 1 (SBF1), this protein has been shown to interact in vitro with SET (Suvar3-9, Enhancer of zeste, trithorax) domains of nuclear proteins. Mutated forms of SBF1 have been shown to be highly oncogenic, and a conserved motif in SBF1 that mediates interactions with SET domains in vitro has been demonstrated to be necessary and sufficient for oncogenic activity. Further, recent studies have demonstrated a role for MTMR5 in spermatogenesis and germ cell differentiation.
