Caspases are produced as zymogens composed of a prodomain plus a large subunit and a small subunit, and require proteolytic processing at internal aspartate residues to generate an active dimeric enzyme. Caspase-13 induces cell death when overexpressed in mammalian cells and is activated by caspase-8, the initiating caspase in the apoptotic cascade.
