The SIRT family of genes are highly conserved from prokaryotes to eukaryotes and are involved in diverse processes including transcriptional regulation, cell cycle progression, DNA damage repair and aging. Sirtuins utilize NAD+ as a substrate to catalyze deacetylation of specific acetylated protein and histones. SIRT1 has been shown to be induced by caloric restriction (CR) in several tissues and SIRT1 mediated deacetylase activity attenuates B-amyloid content in the brain during CR.
HDAC activity (HDAC Fluorimetric Assay -AK-500 from ELS). This assay is performed in two stages. In the first stage, the HDAC(s) acts on the Substrate (10 uM, 50 uM) (total volume of 50 ul). The second stage, is initiated by the addition of 50 ul of developer, stops HDAC activity and produces the fluorescent signal. The samples are read in a microplate reading fluorimeter, capable of excitation at a wavelength in the range 350~380nm and detection of emission light in the range 440~460nm.
