LS-F36238 is a 96-well enzyme-linked immunosorbent assay (ELISA) for the Quantitative detection of Mouse F8 / FVIII / Factor VIII in samples of Plasma, Serum and Tissue Homogenates. It is based upon a Sandwich assay principle and can be used to detect levels of F8 / FVIII / Factor VIII as low as 0.188 nanograms per millilter.

This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- FVIII antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-FVIII antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the FVIII amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration ofFVIII can be calculated.
F8 / FVIII / Factor VIII is coagulation factor VIII, which participates in the intrinsic pathway of blood coagulation; factor VIII is a cofactor for factor IXa which, in the presence of Ca+2 and phospholipids, converts factor X to the activated form Xa. This gene produces two alternatively spliced transcripts. Transcript variant 1 encodes a large glycoprotein, isoform a, which circulates in plasma and associates with von Willebrand factor in a noncovalent complex.
