Purified human MMP-9. Cellular Localization: Cytoplasmic. MW of antigen: 92kD (pro form) and ~86kD (active form).
Recognizes human protein of 92kD and 78kD, under non-reducing conditions, which are identified as pro (latent) and activated forms of matrix metalloproteinase 9 (MMP-9; also known as 92kD collagenase IV or gelatinase B). Also reacts with the complex 200kD) of MMP-9 and tissue inhibitor of metalloproteinases-1 TIMP-1). Shows no cross-reactivity with pro and active forms of MMP-2.

Immunohistochemistry: LS-B2572 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2572 was determined to be 10 ug/ml. Inhibition of MMP-9 Activity: 2-4 ug/ml (order Ab without sodium azide). Immunoprecipitation (Protein G): 2 ug/mg protein lysate (Native verified). Positive control: Conditioned, serum-free medium from (TPA-treated) human fibrosarcoma HT-1080 cells.
