Immunohistochemistry: LS-A4556 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-A4556 was determined to be 20 ug/ml.

CERK, which phosphorylates Cer to produce ceramide 1-phosphate (C1P), was initially described as a Ca2+-stimulated lipid kinase that copurified with brain synaptic vesicles. CERK activity has been reported in HL60 cells, mast cells, and neutrophils. CERK is involved in phagolysosome formation in polymorphonuclear leukocytes and also in liposome fusion. C1P has been reported to have mitogenic effects. More recently, C1P was found to induce arachidonic acid release and prostanoid synthesis.
