ZBTB33 / Kaiso antibody was raised against recombinant N-terminal GST and His-tagged fusion protein corresponding to the central region of murine Kaiso (aa 103-584).
Recognizes Kaiso, Mr 97kD. Species cross-reactivity: Human, mouse, rat, canine, chicken, hamster and simian.
Suitable for use in Western Blot, Immunohistochemistry, Immunoprecipitation and Supershift. Immunohistochemistry: 1 ug/ml gives positive nuclear immunostaining for Kaiso in NIH 3T3 cells fixed with 100% methanol. Western Blot: 0.5-2 ug/ml detects Kaiso in RIPA lysates from 3T3/A31 cells. 3T3/A31 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Kaiso (2 ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. ZBTB33 / Kaiso is a transcriptional regulator with bimodal DNA-binding specificity, which binds to methylated CGCG and also to the non-methylated consensus KAISO-binding site TCCTGCNA. The protein contains an N-terminal POZ/BTB domain and 3 C-terminal zinc finger motifs. It recruits the N-CoR repressor complex to promote histone deacetylation and the formation of repressive chromatin structures in target gene promoters.
