The four human glycoprotein hormones chorionic gonadotropin (CG), luteinizing hormone (LH), follicle stimulating hormone (FSH), and thyroid stimulating hormone (TSH) are dimers consisting of alpha and beta subunits that are associated noncovalently. The alpha subunits of these hormones are identical, however, their beta chains are unique and confer biological specificity. Thyroid stimulating hormone functions in the control of thyroid structure and metabolism.
TSH beta in Human Pituitary. TSH beta was detected in immersion fixed paraffin-embedded sections of human pituitary using Human Thyroid Stimulating Hormone Subunit Beta Monoclonal Antibody at 15 ug/ml overnight at 4°C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic. Tissue was stained using Anti-Mouse HRP-DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm of thyrotrope cells.
Detection of Human TSH beta by Western Blot. Western blot shows purified human pituitary TSH beta (10 ng/lane). PVDF Membrane was probed with 2 ug/ml of Human TSH beta Monoclonal Antibody followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody. A specific band was detected for TSH beta at approximately 25 kD (as indicated). This experiment was conducted under non-reducing conditions.
