Specificity and Use

TAGLN / Transgelin / SM22 antibody was raised against synthetic peptide C-MTGYGRPRQIIS from the C-terminus of human TAGLN / SM22 (NP_003177.2). Percent identity by BLAST analysis: Human, Gibbon, Monkey, Galago, Marmoset, Mouse, Rat, Hamster, Elephant, Panda, Dog, Bat, Bovine, Horse, Pig, Opossum, Xenopus (100%); Stickleback (92%); Turkey, Chicken, Lizard, Catfish, Salmon, Pike, Zebrafish (83%).

Human TAGLN / SM22. NP_003177.2 and NP_001001522.1 represent identical protein.
Immunohistochemistry: LS-B2563 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2563 was determined to be 5 ug/ml.

TAGLN / Transgelin / SM22 is a transformation and shape-change sensitive actin cross-linking/gelling protein found in fibroblasts and smooth muscle. Its expression is down-regulated in many cell lines, and this down-regulation may be an early and sensitive marker for the onset of transformation. A functional role of this protein is unclear. Two transcript variants encoding the same protein have been found for this gene.
