Specificity and Use

RIF1 antibody was raised against synthetic peptide C-RNLQSRWRSPSHEN from the C-terminus of human RIF1 (NP_060621.3; NP_001171134.1). Percent identity by BLAST analysis: Human, Chimpanzee, Orangutan, Gibbon, Monkey, Mouse, Xenopus (93%); Galago, Rat, Panda, Bovine, Dog, Porcine, Guinea pig (86%).

Human RIF1. This antibody is expected to recognize both reported isoforms (NP_060621.3; NP_001171134.1). The immunizing peptide sequence has been optimised for cross-reactiviy in mouse. Reported variants represent identical protein: NP_001171135.1, NP_001171136.1, NP_001171134.1
Peptide ELISA: antibody detection limit dilution 1:8000. Western blot: Approx 270kD band observed in lysates of cell line HeLa and of Mouse Embryonic Stem cells (calculated MW of 274kD according to NP_060621.3). The observed band is suppressed by introduction of Rif1 siRNA to HeLa. Recommended concentration: 1-3 ug/ml.

RIF1 Antibody

Required for checkpoint mediated arrest of cell cycle progression in response to DNA damage during S-phase (the intra-S-phase checkpoint). This checkpoint requires activation of at least 2 parallel pathways by the ATM kinase: one involves the MRN (MRE11A-RAD50-NBS1) complex, while the second requires CHEK2. RIF1 seems to act independently of both these pathways. Seems to play no role in either the G1/S or G2/M DNA damage checkpoints.
