Specificity and Use

Rhodopsin Kinase / GRK1 antibody was raised against rhodopsin Kinase 1a, b (G Protein-Associated Rhodopsin Kinase 1a/b, GRK1a/b).
N-Terminus
Detects G-protein-associated rhodopsin kinase 1a/b (GRK1a/b) from human and bovine tissues. Does not cross-react with other G protein-coupled kinases. Epitope. Recognizes an N-terminal epitope of human GRK1 which is conserved in both GRK1a and GRK1b.
Suitable for use in Western Blot, Immunoprecipitation and Immunofluorescence. Immunofluorescence 1:1000; Immunofluorescence staining of GRK1a/b in bovine retina results in intense staining primarily in cone outer segments, but also in rod outer segments. Weak staining is observed in somata and synaptic terminals of cones and inner segments of rods. Western Blot 1:1000; Detects an ~60kD protein representing GRK 1a/b. Detects both in vitro expressed splice variants by Western blot but was not able to detect significant amounts of GRK1b from human retina.

The G protein-coupled receptor kinases (GRKs) are important enzymes in the desensitization of activated G protein-coupled receptors (GPCR). Among these enzymes, GRK1 is involved in phototransduction and is the most specialized kinase of the family. GRK1 phosphorylates photoactivated rhodopsin (Rho*), initiating steps in its deactivation. GRK1 has been shown to be essential for normal deactivation of cone phototransduction.
