Immunohistochemical analysis of RAR beta staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with hematoxylin and mounted with DPX.

About RARB / RAR Beta
Receptor for retinoic acid. Retinoic acid receptors bind as heterodimers to their target response elements in response to their ligands, all-trans or 9-cis retinoic acid, and regulate gene expression in various biological processes. The RXR/RAR heterodimers bind to the retinoic acid response elements (RARE) composed of tandem 5'-AGGTCA-3' sites known as DR1-DR5. In the absence or presence of hormone ligand, acts mainly as an activator of gene expression due to weak binding to corepressors. P10826 NM_000965 NP_000956.2

Immunogen
RARB / RAR Beta antibody was raised against synthetic 13 amino acid peptide from C-terminus of human RARB. Percent identity with other species by BLAST analysis: Human, Gorilla, Gibbon, Monkey, Marmoset, Bat, Horse, Pig, Opossum (100%); Dog, Turkey, Chicken (92%); Mouse, Panda (85%).

Human RARB. BLAST analysis of the peptide immunogen showed no homology with other human proteins.
Usage
Immunohistochemistry: LS-A6086 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-A6086 was determined to be 10 ug/ml.

Storage
Aliquot and store undiluted at -20°C or below for up to 1 year. Can be stored undiluted at 4°C for up to 1 month. Avoid freeze-thaw cycles.
