Specificity and Use

PPIF / Cyclophilin F antibody was raised against synthetic peptide located between aa92-141 of human PPIF (P30405, NP_005720). Percent identity by BLAST analysis: Human, Chimpanzee, Gibbon, Monkey, Galago, Mouse, Rat, Hamster, Elephant, Panda, Dog, Bovine, Bat, Rabbit, Horse, Pig, Opossum, Guinea pig, Turkey, Zebra finch, Chicken, Xenopus (100%); Gorilla, Marmoset, Platypus, Frog, Catfish, Salmon, Smelt, Stickleback, Pike, Zebrafish (92%); Blood fluke (91%); Drosophila (85%).
Human PPIF / Cyclophilin F
ELISA titer using peptide based assay: 1:62500. Western Blot: Suggested dilution at 5.0 ug/ml in 5% skim milk / PBS buffer, and HRP conjugated anti-Rabbit IgG should be diluted in 1:50000 - 100000 as second antibody.

PPIases accelerate the folding of proteins. It catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides. Involved in regulation of the mitochondrial permeability transition pore (mPTP). It is proposed that its association with the mPTP is masking a binding site for inhibiting inorganic phosphate (Pi) and promotes the open probability of the mPTP leading to apoptosis or necrosis; the requirement of the PPIase activity for this function is debated.
