Specificity and Use

NFKBIA / IKB Alpha / IKBA antibody was raised against synthetic peptide from N-Terminus of human NFKBIA (P25963, NP_065390). Percent identity by BLAST analysis: Human, Chimpanzee, Gorilla, Gibbon, Monkey (100%); Elephant, Guinea pig (84%).

Immunohistochemistry: LS-B1550 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B1550 was determined to be 5 ug/ml.

Inhibits the activity of dimeric NF-kappa-B/REL complexes by trapping REL dimers in the cytoplasm through masking of their nuclear localization signals. On cellular stimulation by immune and proinflammatory responses, becomes phosphorylated promoting ubiquitination and degradation, enabling the dimeric RELA to translocate to the nucleus and activate transcription.
