Specificity and Use

LPIN3 / Lipin-3 antibody was raised against e. coli-derived recombinant mouse Lipin 3 (aa428-529).
Selected for its ability to recognize Lipin 3 in the applications listed. In direct ELISAs and Western Blots, this antibody shows ~50% cross-reactivity with rhLipin 3 and less than 5% cross-reactivity with rhLipin 1 and rhLipin 2.
Suitable for use in ELISA, Western Blot, Immunocytochemistry. Western Blot: 0.1-0.2 ug/ml with the appropriate secondary reagents to detect mouse rmLipin 3. The detection limit for rmLipin 3 is approximately 2 ng/lane under non-reducing and reducing conditions. Immunocytochemistry: 10 ug/ml to detect Lipin 3 on CCD-18co normal human colon fibroblasts. Cells were fixed with PBS containing 4% paraformaldehyde for 20 minutes at RT and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA for 45 minutes at RT. After blocking, cells were incubated with diluted primary antibody overnight at 4?C followed by donkey anti-goat IgG at RT in the dark for one hour. Between each step, cells were washed with PBS containing 0.1% BSA. Immunohistochemistry: 10 ug/ml to detect Lipin 3 in cryosections of adult mouse intestine. Tissue was fixed overnight in 4% PFA + 14% picric acid in PBS at 4?C. Sections were blocked with 10% donkey serum and 0.1% Triton in 1% BSA in PBS for 45 minutes at RT. After blocking, sections were incubated with primary antibody for 3 hours at RT followed by donkey anti-goat lgG at RT for 1 hour. Between each step, sections were washed with 0.1% BSA in PBS. Direct ELISA: 0.5-1.0 ug/ml with the appropriate secondary reagents to detect mouse rmLipin 3. The detection limit for rmLipin 3 is approximately 0.05 ng/well.

LPIN3 / Lipin-3 is a member of the lipin family of proteins, and all family members share strong homology in their C-terminal region. This protein is thought to form hetero-oligomers with other lipin family members, while one family member, lipin 1, can also form homo-oligomers. This protein contains conserved motifs for phosphatidate phosphatase 1 (PAP1) activity as well as a domain that interacts with a transcriptional co-activator.
