Specificity and Use

LEF1 antibody was raised against synthetic peptide (KLH-coupled) corresponding to residues surrounding Pro82 of human LEF1.
Pro82
Detects endogenous level of total human LEF1 protein. Does not recognize the dominant negative forms of LEF1 generated by an alternative promoter. Species cross-reactivity: Mouse and rat.
Suitable for use in Western Blot, Immunoprecipitation, Flow Cytometry, Immunofluorescence and Immunocytochemistry. Other applications not tested.Recommended Dilution:Western Blot: 1:1000. Incubate membrane with diluted antibody in 1X TBS, 5% BSA, 0.1% Tween-20 at 4ºC with gentle shaking, overnight.Immunoprecipitation: 1:50Flow Cytometry: 1:200Immunofluorescence (IF-IC): 1:100Optimal dilutions to be determined by the researcher.

Packaging

10 mM sodium HEPES, pH 7.5, 150 mM sodium chloride, 100 ug/ml BSA, 50% glycerol, less than 0.02% sodium azide.

Participates in the Wnt signaling pathway. Activates transcription of target genes in the presence of CTNNB1 and EP300. May play a role in hair cell differentiation and follicle morphogenesis. TLE1, TLE2, TLE3 and TLE4 repress transactivation mediated by LEF1 and CTNNB1. Regulates T-cell receptor alpha enhancer function. Binds DNA in a sequence-specific manner. PIAG antagonizes both Wnt-dependent and Wnt-independent activation by LEF1 (By similarity).
