Specifications

Description
MUM antibody LS-C37156 is an unconjugated sheep polyclonal antibody to mouse MUM (Latexin) (aa2-222). Validated for ELISA, IP, Neut and WB.
Target
Mouse Latexin / MUM
Synonyms
LXN | ECI | MUM | Protein MUM | TCI | Latexin
Host
Sheep
Reactivity
Mouse (tested or 100% immunogen sequence identity)
Clonality
IgG Polyclonal
Conjugations
Unconjugated
Purification
Affinity purified
Modifications
Unmodified
Immunogen
E. coli-derived rmLatexin (aa 2-222).
Epitope
aa2-222
Specificity
Selected for its ability to recognize mouse Latexin in direct ELISAs and Western Blots. In these formats, this antibody shows ~40% cross-reactivity with rhLatexin.
Applications
  • Western blot
  • Immunoprecipitation
  • ELISA
  • Neutralization
Usage
Western Blot: This antibody can be used at 0.1-0.2 ug/ml with the appropriate secondary reagents to detect mouse Latexin. The detection limit for rmLatexin is approximately 5 ng/lane and 0.5 ng/lane under non-reducing and reducing conditions, respectively. Immunoprecipitation: This antibody has been used to immunoprecipitate rmLatexin from conditioned media of transfected NS0 cells. Neutralization: This antibody has been used to neutralize the inhibitory activity of rmLatexin against rmCPA. The antibody was preincubated with the inhibitor at different molar ratios at room temperature for 30 minutes. The resulting mixtures were then incubated with the enzyme at room temperature for 60 minutes. The enzyme was assayed with a peptide substrate. 50% of the proteolytic activity was restored by the antibody at 4.4 ug/ml (IC50) under conditions in which 0.1 ug/ml of the inhibitor, 0.1 ug/ml of the enzyme, and 100 M of the substrate were present. Considering the molecular masses of the inhibitor (26kD) and the antibody (150kD), IC50 was achieved at approximately 8:1 molar ratio of the antibody to the inhibitor. Direct ELISA: This antibody can be used at 0.5-1.0 ug/ml with the appropriate secondary reagents to detect mouse Latexin. The detection limit for rmLatexin is approximately 0.3 ng/well.
