Specificity and Use

HMOX1 / HO-1 antibody was raised against synthetic peptide with the amino acid sequence DLSEALKEATKEVHC corresponding to residues 12-25 of human heme oxygenase 1. Percent identity by BLAST analysis: Human, Gorilla, Orangutan, Gibbon, Monkey, Marmoset, Mouse, Rat, Hamster, Elephant, Panda, Dog, Bat, Bovine, Pig, Opossum, Platypus, Xenopus (100%); Turkey, Chicken, Lizard (93%).
Recognizes human heme oxygenase-1 (HO-1), a 288 amino acid enzyme with a crucial role in heme catabolism, cleaving the heme ring to form biliverdin which is further processed to form bilirubin by a reductase enzyme. HO-1 acitivity is highest in the spleen where it is involved in the processing of senescent erythrocytes. HO-1expression can be increased during cellular stress, including heat shock. Baseline levels of expression are high in certain tissues, including spleen and liver. Enriched enzyme preparations are recommended for experimental analysis.
Western Blot: Heme oxygenase 1 demonstrates a single band of approximately 32kD. Immunohistology: LS-C11351 reacts with human spleen, kidney and airway biopsies, as well as pig arteries.

Heme oxygenase, an essential enzyme in heme catabolism, cleaves heme to form biliverdin, which is subsequently converted to bilirubin by biliverdin reductase, and carbon monoxide, a putative neurotransmitter. Heme oxygenase activity is induced by its substrate heme and by various nonheme substances. Heme oxygenase occurs as 2 isozymes, an inducible heme oxygenase-1 and a constitutive heme oxygenase-2. HMOX1 and HMOX2 belong to the heme oxygenase family.
