Specificity and Use

NS0-derived rhGranzyme A.
Selected for its ability to detect human Granzyme A in direct ELISAs and Western Blots. In these applications, this antibody shows no cross-reactivity with rhGranzyme B, rhGranzyme H, rmGranzyme D or rmGranzyme G.
Western Blot: This antibody can be used at 1-2 ug/ml with the appropriate secondary reagents to detect human Granzyme A. Using a colorimetric detection system, the detection limit for rhGranzyme A is approximately 5 ng/lane and 25 ng/lane under non-reducing and reducing conditions, respectively. Chemiluminescent detection will increase sensitivity by 5 to 50 fold. Direct ELISA: This antibody can be used at 0.5-1.0 ug/ml with the appropriate secondary reagents to detect human Granzyme A. The detection limit for rhGranzyme A is approximately 7 ng/well.

Abundant protease in the cytosolic granules of cytotoxic T-cells and NK-cells which activates caspase-independent cell death with morphological features of apoptosis when delivered into the target cell through the immunological synapse. It cleaves after Lys or Arg. Cleaves APEX1 after 'Lys-31' and destroys its oxidative repair activity.
