Services
  • Immunohistochemistry Services

  • Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

  • TCR Screening Services

  • Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Immunohistochemistry: LS-B3636 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B3636 was determined to be 10 ug/ml.

Packaging

About FASN / Fatty Acid Synthase

P49327 NM_004104 NP_004095.4

FASN Antibody, Fatty acid synthase Antibody, OA-519 Antibody, SDR27X1 Antibody

The enzyme encoded by this gene is a multifunctional protein. Its main function is to catalyze the synthesis of palmitate from acetyl-CoA and malonyl-CoA, in the presence of NADPH, into long-chain saturated fatty acids. In some cancer cell lines, this protein has been found to be fused with estrogen receptor-alpha (ER-alpha), in which the N-terminus of FAS is fused in-frame with the C-terminus of ER-alpha.

Immunohistochemistry

Immunohistochemistry
Anti-Fatty Acid Synthase antibody IHC of human prostate. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody LS-B3636 concentration 10 ug/ml.

Immunohistochemistry

Immunohistochemistry
Anti-Fatty Acid Synthase antibody IHC of human breast. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody LS-B3636 concentration 10 ug/ml.

Immunofluorescence

Immunofluorescence
Confocal immunofluorescence of FASN Antibody with HeLa cell followed by Alexa Fluor 488-conjugated goat anti-rabbit lgG (green). DAPI was used to stain the cell nuclear (blue).

Immunofluorescence

Immunofluorescence
Fluorescent confocal image of HepG2 cells stained with FASN antibody. HepG2 cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated FASN primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 ug/ml, 5 min). Note the highly specific localization of the FASN mainly to the mainly to the cytoplasm, supported by Human Protein Atlas Data (http://www.proteinatlas.org/ENSG00000169710).

Western blot

Western blot
Western blot of FASN (arrow) using rabbit polyclonal FASN Antibody. 293 cell lysates (2 ug/lane) either nontransfected (Lane 1) or transiently transfected with the FASN gene (Lane 2) (Origene Technologies).

Western blot

Western blot
Western blot of FASN Antibody in Ramos cell line lysates (35 ug/lane). FASN (arrow) was detected using the purified antibody.

Flow Cytometry

Flow Cytometry
FASN Antibody flow cytometry of HeLa cells (right histogram) compared to a negative control cell (left histogram). FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.
