Specificity and Use

EDNRB / Endothelin B Receptor antibody was raised against keyhole limpet hemocyanin (KLH) conjugated C-terminal peptide KANDHGYDNFRSSNN corresponding to amino acids 424 to 437 of rat endothelin B receptor. Percent identity by BLAST analysis: Human, Chimpanzee, Gorilla, Orangutan, Gibbon, Monkey, Galago, Marmoset, Mouse, Rat, Ferret, Sheep, Goat, Hamster, Elephant, Panda, Cat, Bovine, Dog, Horse, Rabbit, Pig, Opossum, Zebra finch, Chicken, Platypus (93%); Lizard (87%).
Recognizes an epitope within the C-terminal (CT) region of endothelin B receptor (ET-B), a widely distributed transmembrane protein and member of the G-protein coupled receptor 1 family, which acts as a non-specific receptor for endothelin 1, 2, and 3, and mediates its action by G protein association and activation of a phosphatidylinositol-calcium second messenger system. AAR43 recognizes epitopes on the cytoplasmic side of the receptor and therefore the cell membrane must be permeable to the antibody. AAR43 does not block the binding of endothelins. AAR43 is reported as suitable for use in immunocytochemistry on acetone fixed cells.

The Endothelin Receptor ETB is expressed in most tissues. Mutations of the receptor gene are responsible for Hirschsprung disease type 2, a multigenic disorder with phenotypes such as bicolored irides, megacolon, hypopigmentation, and hearing loss. The ETB receptor has similar affinity for all three endothelins and activates a phosphatidylinositol-calcium second messenger system.
