Specificity and Use

CREB1 / CREB antibody was raised against proprietary immunogen based on the amino acid sequence containing phospho-serine corresponding to residue 133 of human CREB.
Recognizes phospho-CREB (Ser133), Mr 43kD. Species cross-reactivity: Rat and mouse. Broad species cross-reactivity expected, based on sequence homology.
Suitable for use in Western Blot. Western Blot: 1:1000 to 1:4000. Detects phospho-CREB (Ser133) in RIPA lysates from forskolin treated rat PC-12 and mouse 3T3/A31 cells. Lysates from untreated (Lanes 1 and 3) and forskolin treated (Lanes 2 and 4) PC-12 cells were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho-CREB (Ser133) (1:4000 dilution, Lanes 1 and 2) or anti-CREB (1:2000 dilution, Lanes 3 and 4). Proteins were visualized using a goat antimouse (Lanes 1and 2) or goat antirabbit (Lanes 3 and 4) secondary antibody conjugated to HRP and a chemiluminescence detection system.


0.1 M Tris-glycine, pH 7.4, 0.15 M sodium chloride, 0.05% sodium azide, 30% glycerol.

CREB (cAMP response element binding protein1), a DNA binding protein, is a member of the leucine zipper family of transcription factors. CREB interaction with DNA requires phosphorylation at Ser 119 by activated cAMP protein kinase A. Once phosphorylated CREB binds to an 8 base enhancer know as the cAMP response element (CRE) containing a conserved core sequence TGACG. CREB also interacts with CBP (CREB binding protein) and RNA polymerase II when phosphorylated on Ser 133.
