Antibody RA3-3A1 binds to an extracellular epitope of B220 (CD45R), the 220 kD variant of the T200, CD45 molecule present on B cells and B cell precursors.

Reagent was tested for binding to ACK-lysed murine splenocytes in FACS. Five x 105 splenocytes per tube were washed and pre incubated with 20 ul of 300 ug/ml Mouse IgG (to block non specific binding), after which they were incubated 45 minutes on ice with 80 ul of anti-mouse B220/Biotin at a concentration of 10 ug/ml. Cells were washed twice and incubated with secondary reagent Streptavidin/PE after which they were washed three times, fixed and analyzed by FACS. A 33% sub population of the cells stained positive with a mean shift of 2.44 log10 fluorescent units when compared to background. Binding was blocked when cells were pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-mouse B220 antibody. The applications listed have been tested for the unconjugated form of this product. Other forms have not been tested.
