Specificity and Use

APOB / Apolipoprotein B antibody was raised against apolipoprotein Type B was isolated from human plasma by density gradient centrifugation followed by HPLC purification.
Typically less than 1% cross reactivity againstother types of apoLipoprotein was detected by ELISA against purified standards. This antibody reacts with human apoLipoprotein B and has negligible cross-reactivity with Type A-I, A-II, C-I, C-II, C-III, E and J apoLipoproteins. Specific cross reaction of anti-apoLipoprotein antibodies with antigens from other species has not been determined. Non-specific cross reaction of anti-apoLipoprotein antibodies with other human serum proteins is negligible
Immunohistochemistry: LS-B3646 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B3646 was determined to be 5 ug/ml.

Apolipoprotein B (ApoB) is the main apolipoprotein of chylomicrons and low density lipoproteins (LDL). The protein occurs in the plasma in 2 main isoforms, apoB-48 and apoB-100. The first is synthesized exclusively by the gut, the second by the liver. The intestinal (B-48) and hepatic (B-100) forms of apoB are coded by a single gene and by a single mRNA transcript larger than 16 kb. The 2 proteins share a common amino terminal sequence.
