SIRPA / CD172a Antibody - 1:100 staining human brain tissue by IHC-P. The tissue was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The tissue was then blocked and incubated with the antibody for 1.5 hours at 22°C. An HRP conjugated goat anti-rabbit antibody was used as the secondary.
SIRPA / CD172a Antibody - Staining HepG2 cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% saponin prior to blocking in 10% serum for 45 min at 37°C. Usage
For western blots: incubate membrane with diluted antibody overnight in 5% w/v milk , 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking.

SIRPA / CD172a Antibody - Immunofluorescent analysis of CD172a staining in THP1 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
